中药艾迪诱导人喉癌细胞Hep-2凋亡的研究

  • 摘要: 目的 研究中药艾迪对人喉癌细胞Hep-2的生长抑制作用及凋亡诱导作用。 方法 四亚基噻唑蓝(MTT)法测定中药艾迪对人喉癌细胞Hep-2生长的抑制作用,荧光显微镜观察人喉癌细胞Hep-2的凋亡状况,流式细胞仪技术(FCM)观察人喉癌Hep-2细胞周期及凋亡,荧光定量PCR法测定人喉癌细胞Hep-2环氧化酶(COX-2)的表达量。 结果 不同浓度的中药艾迪对人喉癌细胞Hep-2生长均有抑制作用,并有明显的时间和剂量依赖性,与空白对照组比较,差异具有统计学意义(P<0.05)。流式细胞仪分析显示中药艾迪能减少G1期的比例,增加G2和S期的比例,并能诱导细胞凋亡。中药艾迪作用人喉癌细胞Hep-2 24h后其环氧化物酶-2的表达量升高,与空白对照组比较,差异具有统计学意义(P<0.05)。 结论 中药艾迪在体外能明显抑制人喉癌细胞Hep-2生长,其机制可能是诱导细胞凋亡。

     

    Abstract: Objective To study the effect of Aidi,a Chinese medicine,on growth and apoptosis of laryngeal cancer cell line Hep-2. Methods Effect of Aidi on growth of Hep-2 cells was detect by MTT assay.Apoptotic Hep-2 cells were observed under a microscope.Cell cycle of Hep-2 cells was detected by flow cytometry(FCM).Expression of epoxidase in Hep-2 cells was detected fluorescence quantitative PCR. Results Aidi at different concentrations inhibited the growth of Hep-2 cells in a significant time-and dose-dependent manner compared with that in controls(P<0.05).FCM showed that Aidi decreased the apoptotic rate of Hep-2 cells at the G1 phase,increased the apoptotic rate of Hep-2 cells at the G2 and S phases and induced the apoptosis of Hep-2 cells.The expression level of epoxidase was higher in Hep-2 cells than in control 24h after treatment with Aidi(P<0.05). Conclusion Aidi inhibits the growth of human Hep-2 cells by inducing apoptosis of them.
