The study on the effective factor of the ocean microbs detection

  • Objective:To provide helpful information on the surveillance of those microbs by studying the optimal culture condition of the ocean microbs.Methodes:Sea water is collected by the sterile sea water colletor, and each 225ml of the sea water is applied to 25ml enrichment broth (10×alkline peptone water,5×alkline peptone water, 1×alkline peptone water,saltphilic enrichment broth,Rappaport Vassiliadis Soya Peptone (RVS)Broth,and nutrient broth.The upabove cultures are cultured on 18℃,25℃ and 35℃ respectively,with an interval of 6 hours. The surface cultures are then inoculated on the responding cultures (part of the cultures are made of sea water and distilled water as a contrast),the grown-up bacteria are indentified with the pure colony.Results:The optimal temperature for the culture of ocean microbs is 35℃; corresponding multification time is between 6 and 12 hours; and the culture time is between 12 and 24 hours;among all peptone water, the multification result is the best with 10×alkline peptone water,and the cultures made by sea water are better than those made by the distilled water. Conclusion:Ocean microbs are saltphilic, cultures with appropriate content of salt are good for the culture of ocean microbs.
