Sox9 gene therapy for intervertebral degenerative disc of animal model

Abstract
Objective: To assess adenoviral delivery vectors expressing Sox9 gene therapy for intervertral degenerative disc of animal model. Methods: The model of intervertebral degenerative disc of rabbit was established by stabbing into the L2-3,L3-4 and L45discs to product a puncture injury using a 16-gauge hypodermic needle and after 24weeks,the intervertebral generative discs were injected into adenoviral delivery vectors expressing Sox9 gene.After 30weeks(6weeks after injecting adenoviral delivery vectors expressing Sox9 Gene) we observed changes of the intervertebral generative discs by MRI and took out the intervertebral generative discs to observe changes of type 2 collagen production by Col2al mRNA by RT-PCR.Results: The intervertebral generative discs were recovered in a great measure on MRI and exogenous Sox9 upregulated Col2al mRNA expression.Conclusions: adenoviral delivery vectors expressing Sox9 gene therapy for intervertral degenerative disc of animal model is effective.
