Micro-lymphocytotoxicity test and PCR-dye assay in detection of HLA-B27:A comparative study

Abstract
Objective To study the application of PCR-dye assay and micro-lymphocytotoxicity test in detecting HLA-B27 by analyzing their advantages and disadvantages. Methods HLA-B27 was detected in 197 patients with ankylosing spongylitis(AS) by PCR-dye assay and micro-lymphocytotoxicity test,respectively.The inconsistent findings in PCR-dye assay and microlymphocytotoxicity test were tested by sequencing. Results Micro-lymphocytotoxicity test showed positive and negative HLAB27 in 150 and 47 out of the 197 patients.PCR-dye assay displayed positive and negative HLA-B27 in 135 and 62 out of the 197 patients(P<0.01).The HLA-B27 detection rate was significantly higher by micro-lymphocytotoxicity test than by PCR-dye assay in 15 patients with different findings(86.7% vs 13.3%,P<0.05). Conclusion The accuracy of PCR-dye assay is better than that of micro-lymphocytotoxicity test in detecting HLA-B27 and is advantageous over micro-lymphocytotoxicity test in sample processing and easier to perform,thus having a bright future in clinical application.
