Correlation of single nucleotide polymorphisms in TLR2 and TLR4 receptors with Acinetobacter baumanii infection

Abstract
Objective Todetermine the correlation of SNP polymorphisms of TLR2 and TLR4 gene with Acinetobacter baumanii infection and provide basis for early prevention of Acinetobacter baumanii infection. Methods Four hundredand twenty- three Acinetobacter baumanii infectedpatients and 385 patients collectedfromthe same wardwere selectedas expertimental groupand control group, respectively. The cand idate SNPs at TLR-2 (rs3804099, rs7656411 and rs76112010) and TLR-4 (rs1927914, rs10759932 and rs11536889) were selectedaccording totheir frequency (> 5%), position, and their associations with infectious diseases reportedpreviously. SNP genotyping was performedusing an improvedmultiplex ligation detection reaction (iMLDR) technique. The relationshipbetween Acinetobacter baumanii infection and SNPs polymorphisms of TLR2 and TLR4 was analyzedby multiple logistic regression model. Results All SNPs were in Hardy-Weinberg equilibrium(HWE). The hyplotype distribution of TLR2 and TLR4 cand idate alleles showednosignificant difference between infectedpatients and controls, while the haplotype GCG of TLR-4 was significantly associatedwith Ab infection (P=0.027). Conclusion Our studies suggest that the twoSNPs (rs1927914 and rs10759932) of TLR-4 may be relatedtothe susceptibility toAb infection.
