Effect of CYP2C9 genetic polymorphism on maintenance warfarin dosage in Chinese: A Metaanalysis

Abstract
Objective To analyze the effect of CYP2C9 genetic polymorphism on maintenance warfarin dosage in Chinese. Methods A meta-analysis of studies about the effect of CYP2C9 genetic polymorphism on maintenance warfarin dosage in Chinese covered in related databases from 2000 to 2011 was performed using Stata10.0 software. Results Sixteen studies involving 2 361 patients were included in the meta-analysis.The wild-type CYP2C9*1,mutant CYP2C9*2 and CYP2C9*3 accounted for 93.99%,0.06%,and 5.95%,respectively,of CYP2C9 alleles in Chinese.The maintenance warfarin dosage for genotypes CYP2C9*1/*3 and CYP2C9*3/*3 was 26.8%(95%CI 18.8%,34.8%) and 26.3%(95%CI 12.6%,39.9%) lower than that of wild-type CYP2C9 *1/*1.Subgroup analysis and sensitivity analysis demonstrated that the results of meta-analysis were stable and reliable. Conclusion CYP2C9 genetic polymorphism in Chinese populations is different from that in other races and exerts an important effect on maintenance warfarin dosage.
