ESTABLISHMENT OF HUMAN PULMONARY GIANT CELL CARCINOMA CELL LINE (PLA-801)AND OBSERVATIONS ON ITS BIOLOGIC PROPERTIES

Abstract
A cell line designated as PLA-801 has been derived from the pleural fluid of a patient with giant cell carcinoma of the lung. It has been transferred for 110 passages in a period of one year and three months. The course of establishment and some of its biologic properties are presented. The anchorage-dependent cells grew into monolayer growth within 48 hours and the majority of the cells were arranged in a pattern of epithelium, The first passage was on the 28th day and then subculture was made once every week. The growing cells of one bottle was transfered into 2-3 bottles. and the medium was changed 3 times every week. Biologic properties of the PLA-801 cell line: Microscopically the cells were charac-terized by pleomorphism and epithelium-like pattern with some prominent bizarre giant cells as well as multinucleated giant cells with abnormal mitotic figures. A large number of cells appeared as round cells. some of these cells crowded into groups. and these forms could be observed frequently on the 3rd day of passage. The growth curve of the cell line was determined from the 60th and 105th passage, of which the cell number was counted daily for a week. The growth curve of the both passages was similar. The doubling time was 29 hours. The mitotic index was determined by counting 2000 cultured cells daily from the 1st to 7th day. Chromosome analysis of cells was performed by observing 100 metaphase of mitotic division. and it was found that the number of chromosome varied in different cells. being 70-79. This cell line may be considered to be hypertriploid. Other observations. such as Con-A agglutination. efficiency determination of ectopic gonadotropin. animal transplantation and electron microscopic examination. are briefly discussed.
