Radioimmunoiimaging of human colon carcinoma xenogafts in nude mice by 111ln-labelling monoclonal antibody

  • A nude mouse model bearing human colon carcinoma xenograft from fresh surgical specimens of patients was established. The an ti-colon carcinoma monoclonal antibody 2C10 was labelled with 111In using chelating agent DTPA. The labelled antibody was injected into the nude mice with tumour xenograft via peritonea. The tissue distribution of labelled antibody was studied and whole body scintigraphies with a r camera were obtained at different times. 96 hours after administration, the tumour to muscle ratio was 10.48; tumour to blood ratio 3.94; and tumour to liver ratio 1.58. There were significant differences in T/N ratio between the experimental and control group. The result of our experiment suggested the possibility that 111In-labelling McAb 2C10 can serve as a tracing agent in human tumour radioimmunodiagnosis.
