Abstract
Objective: To identify molecular markers correlated with the differentiation of CRC.Methods: Affymetrix oligonucleotide microarrays HG-U133 were used to monitor gene expression on CRC and paired normal mucosa tissue.The tissue samples of CRC were classified by Tclass(Tumor classification system) in molecular level.The expression difference of candidate genes in different tissues was confirmed by real time FQ-RT-PCR(real time fluorescent quantitative reverse transcription polymerase chain reaction).Results: 8 genes & ESTs with 100% classification accuracy were obtained by Tclass,in which C8orf4 gene has showed lower mRNA expression in the CRC tissue than in paired normal tissue by multifold statistical methods.The normalized expression levels of C8orf4 were significantly different among CRC with different differentiation grades which was confirmed by real time FQ-RT-PCR,and have linear correlation with its expression in GeneChip.Conclusion: C8orf4 is closely correlated with the differentiation of colorectal cancer and can be regarded as candidate molecular marker.Instestinal tumor;PCR
