The response of the vessel to the injury:A canine restenotic model after PTCAestablished

Abstract
The efforts to reduce or eliminate restenosis after PTCA have been hampered by the lack of an ac-curate animal restenotic model with substational proliferation,This study was thus designed to determine theprobabilities and the mechanisms of developing a practical proliferative canine model by using deep injury tothe media of the coronary and femoral artery.Stainless steel wire coils were delivered percutaneously to the14 coronary and 24 femoral arteries in 26 canines with a oversized,high-pressure(1 215.9~141 8.5 kPa )balloon and were left in situ from 1,7,14,30,60 to 90 days. Histopathologic examinations was made afterangiograghy.The histologic examination showed that the substantional neointimal proliferation occured in 4~12 weeks with a lulninal narrow area more than 50%at least;the severity of the vessel injury-the mean in-jury score strongly correlated with neointimal thickness(r=0.862)and percent area setenosis(P<0.05),and the histopathologic results in femoral arteries were the same as those in coronary arteries.Our results in-dicated that this model is a practical and inexpensive one,and the histopathologic features of the proliferativeresponse were identical to those observed in human caces of restenosis after angioplasty.
