Study on preparation of adult human cardiac myosin heavy chains

Objective: To explore an efficient method for the purification of human cardiac myosin heavy chain （MHC） so as to provide the antigen for making its antibody.Methods: Adult human cardiac MHC was purified by a modefied method,its purity and immunological properties was identified by a discontinous sodium dodecyl polyacrylamide slab gel electrophoresis （SDS PAGE）system and Western blot.Results: （1）the electrophoretogramof the purified MHC showed a typical pattern of one band representing MHC; （2）Western blot showed a band at 200 ku corresponding to the electrophoretogram MHC band.Conclusion: The modefied method of purifying human cardiac MHC is practicable and efficient;the MHC prepared by the method here has a high purity,therefore,it may be used as the antigen to produce MHC specific antibody
