Effect of leptin on PDX-1 gene expression in isolated rat islets

Objective: To investigate the effect of different concentrations of leptin on PDX-1 gene expression in isolated rat islets.Methods:Leptin was added to isolated islets at final concentrations of 0,10,25,50,102,103 ng·mL^-1 to media containing 2.2 mmol·L^-1（low）,5.5 mmol·L^-1（normal） and 11.1 mmol·L^-1（high） glucose.All islets samples were harvested after incubation for 24 h,and total RNA was extracted for a single series for PDX-1 RT-PCR.Results: There was no change in PDX-1 gene expression with different concentrations of leptin with glucose of 2.2 mmol·L^-1,5.5 mmol·L^-1 and 11.1 mmol·L^-1.Conclusion: Leptin has no significant effect on PDX-1 gene expression.
