Analysing ultrastructure characteristic in the ruthenium red staining on the rats with IPF

  • Objective: To investigate ultrastructure characteristic in the ruthenium red staining on the rats with IPF.(Methods: 180) male SD rats were randomly divided into six groups: making a model by injecting BLM in trachea,treating by high-dosage,medium-dosage and low-dosage medicine.each group are killed respectively on 7th day,14th day and 28th day.the ultrastructural change by ruthenium red staining were observed.Results: Collagen fiber is in focus around basement menbrane and cell.Cell menbrane and organelle is more clear,there are pathological change of AT-Ⅱ on 7th day,14th day and 28th day in differ phase.we can see a binds of collagen fiber espesially between basement menbrane and cell.14th day basement menbrane outboard there are pucker and distortion or in faith.There are ruthenium red staining sediment in the basement menbrane,blood-air barrier becomes thicker.28th day basement menbrane becomes asymmetry and integrality is destroyed.Forecollagen fiber are monifolding and rough endoplasmic reticulums in the cell are expanding,collagen fiber accumulates in interstitial and arrange cross.(Conclusion: Basement) menbrane and collagen fiber round cell may be revealed more clear in the ruthenium red staining on the rats with IPF.it is more convenience observing distributing of collagen fiber and thick of basement menbrane.staining;idiopathic pulmonary fibrosis;collagen;microtomy
