Clinical and hemodynamic observation of single dose Amrinone given intravenously in patients with heart failure

Clinical and hemodynamic studies were performed in patients with heart failure with a purpose of evaluating the immediate effects of Amrinone. 10 patients aged 42—63 years received one dose of amrinone, 1 mg/kg intravenously for evaluation of the shortterm therapeutic effects in congestive heart failure （5 coronary, 2 hypertensive heart disease and 3 congestive cardiomyopathy）.The results showed that the cardiac output increased 32.8% （P＜0.01）, PCWP declined 45.3% （P＜0.001） No sigificant changes in HR and Bp were found.A Comparative study of serum and RBC cAMP levels were studied coincidently with hemodynamic monitoring. During treatment only 2 patients developed mild transient thrombocytopenia and recovered one week later.
