Determination of ATP in the rat heart with ischemia reperfusion by ion-pair reversed-phase HPLC

Abstract
Objective: To study the change alteration of ATP in the rat heart with ischemia reperfusion injury by lon-pair reversed-phase HPLC. Methods: Ion-pair reversed-phase HPLC methods was developed for determination of ATP.The solvent consist of 0.1 mmol·L-1 KH2PO4;8 mmol·L-1 tetrabutylammonium hydrogen.The separation was monitored at 259nm.(Results: The) calibration curve showed good linearity(r=0.998).The coefficients of variation(C.V) were estimated from(repeated) determinations of standard ATP.The intraassay C.V varied from 1.91% to 5.78%.The recovery was between 97.25% and 100.05%. Conclusions: This methods is precise,it can be used for determination of ATP in the rat heart.
