Experimental study about acute DIC

  • Experimental disseminated intravascular coagulation(DIC)was induced by sustained infusion of thrombin and EACA into the ear vein in rabbits.The dynamic changes of coagulation factors, Prostaglandin metabolism,and the condition of microcirculation were determined with reference to various parameters.The DIC thus induced showed a close resemblance to human DIC as judged from such changes as an elevation in the rate of positive reaction in 3P test,prolongation of PT and KPTT,the decrease in fibrinogen and platelet count,and the presence of numerous microthrombi in the pulmonary capillaries and renal glomeruli. It can be seen that the platelet aggregation induced by adenosine diphosphate(ADP)had an elevation first and then a decrease and that platelet adhesion abated during the formation of DIC. The concentration of TXB2 and that of 6-keto-PGF were increased but the concentration of MDA had no significant changes.The speed of the blood flow in the microcirculation became slower. Following treatment with anisodamine,the rate of positive reaction of 3P was delayed,PT was kept in normal range,consumption of fibrinogen was ameliorated,platelet aggregation induced by ADP was inhibited,the concentrations of TXB2 and MDA were significantly lower as compared with those in the control group,but the quantity of 6-keto-PGF showed no distinct changes.
