Effect of bcl-2 antisense oligodeoxynucleotides on human renal cell carcinoma cells growth

  • Objective:To evaluate the effect and dose-response relationship of antisense oligodeoxynucleotides(ODNs) on inhibition of renal cell carcinoma growth in vitro. Methods: Design and synthesize antisense targeting various regions of the bcl-2 mRNA without the homology of the human genes. ODNs were delivered to human renal cell carcinoma cells in the form of complexes with liposome. The cytotoxicity of ODNs in RCC cells was determined by measurement of cell viability by use colorimetric assay.Results: Treatment with transfected antisense bcl-2 ODNs inhibited the growth of all tested RCC cells, but Liposome or ODNs alone. Inhibition by antisense bcl-2 ODNs of ACHN cells was dose dependent. Conclusions: Transfected ODNs effectively reduced the viability of many kinds of RCC cell in dose-dependent manner.
