Pharmacokinetic investigation of sisomicin in Chinese

Sisomicin in doses of 1 mg/kg was administered intravenously to 13 healthy volunteers, by the of use agar diffusion method. The sisomicin concentrations in the serum and urine were determined using bacilli subtilis 63531 as the test bacteria. The blood samples were collected 0.1, 0.5, 1.0, 2.0, 4.0, 8.0, 12.0, 24.0, hours the administration of drug, while the urine samples were collected after 0-2, 2-4, 8-12, 12-24 hours. Pharmacokinetic parameters were calculated by the Tang’s method of damping nonlinear least squares. The beta phase was B=4.02±1.01, beta= 0.36±0.07 and the alpha phase was A=10.17±5.39, alpha=7.55±7.80. This concentration calculative formular is C=10.17e^-5.391+4.02e^0.361. These results were obtoined in seven healthy volunteers of middle age. And they may be used in clinical monitoring of drug therapy, particularly in old patients.
