Foundation of acute graft-versus-host disease model in murine haploidentical stem cell transplantation and exploration of secondary cytopenia

  • Objective: To find acute graft-versus-host disease model in murine haploidentical stem cell transplantation and to explore secondary cytopenia after transplantation.Methods: C57BL/6N mice were donor,BCF1 mice(C57BL/6N×BALB/c) were recipient.Recipient mice were divided into irradiation group(R)、 irradiation add inoculating bone marrow cell group(B)、 irradiation add inoculating bone marrow cell add splenocyte group(S).After transplantation,mice were checked daily for the appearance of aGVHD symptoms such as hunched posture,ruffled fur,diarrhea,and survival.Blood routine test was done twice weekly,chimerism was detected by FACS,and Liver,skin,intestine,and spleen were biopsied for histopathological examination.Results: S group showed reduction of weight,hunched posture,ruffled fur and death.aGVHD was verified by histopathological examination.Secondary cytopenia after transplantation occurred in S group,while chimerism was more than 90% at the same time.Conclusion: aGVHD model in murine haploidentical stem cell transplantation can be founded when irradiation dose of 9.5 Gy,inoculated with 107 bone marrow cell and 3×107 or 4×107 or 5×107 splenocyte,and aGVHD was main reason of Secondary cytopenia in S group.
