Phenotypes of CD25,CD29 and HLA-DR cells before and after in vitro amplification from human peripheral blood lymphocytes

  • Objective To study the phenotypes of CD25,CD29,and HLA-DR cells before and after in vitro amplification from peripheral blood lymphocytes of 35 malignant tumor patients. Methods Lymphocytes and mononuclear cells were induced and amplified from peripheral blood of malignant tumor patients. Percentages of CD25+,CD29+,HLA-DR+,CD4+CD25+,CD4+CD29+,CD3+HLA-DR+,CD3+HLA-DR-,CD4+HLA-DR+,CD4+HLA-DR-,CD8+HLA-DR+,and CD8+HLA-DR-cells in effector cells were calculated by flow cytometry,and compared with their percentages before in vitro amplification. Results After amplification,the percentages of CD25+,CD29+,CD3+HLA-DR-,CD8+HLA-DR+ and CD8+HLA-DR-cells were significantly increased(P<0.01), while the percentages of CD4+CD25+,CD4+CD29+,CD4+HLA-DR+ and CD4+HLA-DR-cells were obviously decreased(P<0.01). No significant change was found in the percentages of HLA-DR+ and CD3+HLA-DR+ cells(P>0.05). Conclusion After amplification,the percentage of CD4+CD25+ regulatory T cells(Treg) is obviously decreased while the percentage of active T cells does not change significantly before and after amplification.
