Expression of L-CPT1 mRNA in liver of scalded rats with wound sepsis

Abstract
Objective: To investigate the expression of liver carnitine palmitoyltransferase 1 (L-CPT 1) mRNA in liver of scalded rats with wound sepsis. Methods: 60 male Wistar rats were scalded resulting in 30% TBSA full-thickness burn and then randomly divided into wound sepsis group and control group. Pseudomonas aeruginosa (109 /ml) was inoculated to the rats’ wounds of sepsis group. The quantity of bacteria in the subeschar tissue was mensurated to confirmed that the rat model of burn wound sepsis was established successfully. Fresh liver tissues were obtained. Total RNA of liver was isolated with Trizol and the levels of L-CPT 1 were measured by reverse transcription polymerase chain reaction (RT-PCR). Results: Compared with control group, L-CPT 1 mRNA levels of sepsis group were significantly decreased in liver during 24-96h post scalding (P<0.05). Conclusions: Burn wound sepsis can decrease the expression of L-CPT 1 mRNA in liver, which might decreased the entry of long-chain fatty acids into the mitochondrial matrix, and involved in the development of lipid metabolism disturbance.
