Effect of propofol on prostacyclin and thromboxane during hemorrhagic shock and resuscitation in monkeys

  • Objective: To investigate the effect of propofol on the plasma concentrations of prostacyclin(PGI2) and thromboxane A2(TXA2). Methods: Twelve monkeys with hemorrhagic shock were devided randomly into two groups.The monkeys in propofol group received propofol at plasma concentration 8mg/L by target control infusion,while monkeys in control group received normal saline in the same.Venous blood were collected to determine plasma prostacyclin (PGI2) and thromboxaneA2(TXA2)concentrations by radio-immunoassay before bleeding, 2 hours after hemorrhagic shock and 2 hours after resuscitation. (Results: Compared) with control group’s,the TXA2 level of propofol group was significantly lower at 2 hours after resuscitation((P=0.0001)), and the ratio of PGI2/TXA2 of propofol group was higher(P=0.0001). Conclusion: These findings indicate that propofol can decrease the plasma concentration of TXA2 in hemorrhagic shock and resuscitation monkeys,which may be beneficial to control the development of hemorrhagic shock.
