Different expression of ENaCs mRNA subunits in spontaneous hypertension rats and Wistar rats

Abstract
Objective To verify the different expressions of ENaCs mRNA subunits in tissues of spontaneous hypertension rats(SHR) and Wistar rats and investigate the role of mechanosensitive ion channels in arterial baroreceptor resetting. Methods Left carotid sinuses,aortic arches,and vagus nerves in 6 SHR and 6 Wistar rats were separated.Total RNA in these tissues was extracted with the guanidinium isothiocyanate-chloroform method.FQ-PCR was developed to measure the changes in expressions of α,β,and γ ENaC mRNA in tissues of SHR and Wistar rats. Results The expression level of ENaCs was higher in tissues of Wistar rats with normal blood pressure than in tissues of SHR,especially in carotid sinuses. Conclusion Down regulation of ENaCs expression in hypertensive condition indicates that mechanosensitive ion channel may play a role in the baroreflex resetting.
