Detection of differential peptides in serum of rat anti-Thy-1 model by Seldi-Tof-MS

  • Objective To detect the differential peptides in serum of rat anti-Thy1 nephritis model during their proliferation in mesentery by Seldi-Tof-MS in order to find mesangial cell proliferation-related biomarkers. Methods Twelve male Wistar rats weighing 200g were divided into control group and model group.Rats in control group were injected with physiological saline and rats in model group were injected with anti-Thy1 antibody through tail vein.Animals were killed 7 days later.Periodic acid-Schiff(PAS) staining was used to detect pathologic changes in the two groups and Seldi-Tof-MS was used to detect differential peptides in serum of rat anti-Thy1 nephritis model. Results Significant proliferation of mesangial cells and extracellular matrix accumulation were observed in model group 7 days later.Two differential peptides were found in model group with a high expression level of M/Z 2 746ku peptide and a low expression level of M/Z 7 399ku(P<0.01). Conclusion Two differential peptides can be found in serum of anti-Thy1 nephritis model using Seldi-Tof-MS,which may be the mesangial cell proliferation-related biomarkers.
