Studies on toxicity and clinical application of cryoprotective agent dimethyl sulfoxide (DMSO)

The effects of DMSO on the viability of bone marrow cells, fetal liver cells and lymphocytes at 4℃ and room temperature, the LD₅₀ dose of DMSO in mice and dosage of DMSO used in patients are reported. It was shown that in comparison with the preservation at 4℃, cell viability and CFU-GM yield were markedly decreased when preserved at room temperature The LD₅₀ dose of DMSO by single intravenous injection in mice was 3.0364±0.8709mg/g body weight. Our experimental studies in mice and dogs, and clinical application, revealed that unwashed and diluted cryopreserved bone marrow and fetal liver cell suspension may be successfully infused to animals and patients without any harmful side-effect.
