Expressive analysis of human gastric cancer-associated susceptible gene in Situ

Abstract
Objective: To further study the expression of the human gastric cancer-associated susceptible gene cDNA W41 in gastric cancer tissues. Methods: C-PRINS technology are applied to detect the expression of W41in gastric cancer and normal(tissues),in the same time bioepiderm with HRP(horseradish peroxidase) labeling and DAB chromogenic method were employed so as to increase sensibility of detecting W41 mRNA on paraffin section.Image analysis sofeware was used to quantitate the expression of W41,and mean OD was used as the quantitative index. Results: In Situ C-PRINS showed that W41 expressed significant difference in gastric cancer,intestinal metaplasic(IM),none-IM paratumor tissues and normal gastric mucosas by F test((F=8.810,) P=0.001).W41 expression existed no difference between tumor and paratumor tissue(P>0.05),while its expression was significantly lower in normal tissue than in tumor and paratumor tissues(P<0.05).there was no difference between(adenocarcinoma) and signet-ring cell carcinoma(t=1.304,P=0.029). Conclusion: W41 presented higher expression in gastric cancer tissue,paratumor tissue than normal tissue,and apparente expression in IM,that suggests W41 is linked to the gastric(carcinogenisis). C-PRINS method is superior to the classic hybridization in situ,with the advantages of stronger signal,easy(manipulation),and time saving.
