Effect of Naloxone on delay of the collapse of mitochondrial membrane potential and improvement of energy metabolism of cultured myocardial cell induced by hypoxia and reoxygenation

Abstract
Objective: To study the effect of Nalonxe on mitochondrial membrane potential(MMP) in cultured myocardial cell after hypoxia-reoxygenation and its possible protection to cardialmyocytes′energy metabolism.Methods: cultured cardiomyocytes from neonate rabbits were divided into three groups(control group,hypoxia group and naloxone group) and the two latter were further divided into three subsections: hypoxia 2 hour,hypoxia 2 h-reoxygen 2 h,hypoxia 2 h-reoxygen 4 h.Then the cells were stained with fluorescent dyes and MMP was assayed by flow cytometry.Results: After hypoxia,MMP of hypoxia group and Naloxone group were lower than the control(P≤0.01).MMP of therapy group was significantly higher than that of hypoxia group on each different time point(P≤0.01).Conclusion: Naloxone might significantly reduce the lossing of MMP of hypoxia-reoxygenation injured cardialmyocytes and improve their energy metabolism.
