The preparation and biochemical analysis of extracts from burned and normal skin of booth human and rat

  • Objective: According to the protocol of Allg wer and his colleagues, we have established a modified method for the preparation of burned and normal skin extracts of both human and rat. Methods: We performed the SDS-PAGE as well as the ultra-violet chromatographic analysis and total protein quantitation of the normal and burned skin extracts of human (HNSE and HBSE) and rat (RNSE and RBSE). With the combination of mice toxic texic tests. Results: We have verified the HBSE as a toxic complex of two protein with molecular weight of 228ku and 138ku. We also verified the HNSE as a non-toxic complex of two proteins with molecular weight of 148ku and 79ku. The RBSE was proved to be a toxic complex of a 218ku protein and a 138ku protein whereas the RNSE was a non-toxic complex of a 99ku protein and a 79ku protein. Conclusion: The burned skin extracts from both human and rat can significantly induce the pathologic changes in certain organs in mice when intra peritoneally injected.
