Protective effects of PTD-BDNF on cerebral ischemia injury in rats

  • Objective To observe the in vitro effects of PTD-BDNF on ischemia reperfusion injury in rats. Methods A rat model of middle cerebral artery occlusion(MCAO) was established by intraluminal filament occlusion with thread ligation. Rats in PTD-BDNF group were intravenously injected with 2mg/kg PTD-BDNF immediately after ischemia reperfusion,and rats in control group were injected with the same volume of normal saline. TTC,HE and TUNEL staining were used to analyze the neuroprotective effect of PTD-BDNF on ischemia reperfusion injury in rats. Results Compared with the control group,the percentage of infarct volume and TUNEL positive rate decreased from 18.20% to 10.12% and from 51.24% to 23.54%,respectively,in PTD-BDNF group(n=4,n=6,P<0.01), implying that PTD-BDNF can significantly decrease the infarction volume and apoptosis ratio,and increase the survival time of neurons in rats with focal cerebral injury induced by ischemia-reperfusion. Conclusion Intravenous PTD-BDNF can protect neurons from injury across the blood brain barrier,reduce ischemia reperfusion injury and improve the function of cranial nerves.
