Abstract
Objective: To study the neutralization effect of anti-human TNF-α monoclonal antibody on the TNF-α in sera of the psoriatic patients. Methods: We used sandwich ELISA to detect the concentrations of TNF-α in sera of healthy volunteers and patients before and after neutralization with antibody. L929 cells were Used to detect the bioactivities of TNF-α in sera of healthy volunteers and patients before and after neutralization with antibody. Results: The concentrations of TNF-α in sera of healthy volunteers and patients were(3.146±6.184) pg/ml and(62.723±38.379) pg/ml respectively;their bioactivities were(6.817±5.086) % and(57.498±9.125)% respectively. After neutralization with antibody, the concentrations of TNF-α of patients were(0.503±1.178)pg/ml, and the bioactivities of TNF-α of patients were(25.378±14.863)%. Conclusions: We can find that the concentrations and bioactivities of TNF-α in sera of patients were elevated, and anti-human TNF-α monoclonal antibody can well neutralize the concentrations and bioactivities of TNF-α in psoriatic patients’ sera.
