Study on relationship between expression of mechanical signal related proteins and validity of skin after cryopreservation

  • Objective:To investigate the changes of integrin β1 and talin in epidermal cells treated by different temperatures.Methods: The independent experiments were performed with samples from the same individual.These samples were preserved by 4 methods: 4 ℃,-20 ℃,-80 ℃,-196 ℃,to be compared with fresh skin.The experiments were carried out 48 hours after preservation.Immunohistochemistry technology was adopted for the examination.Then the histological structure of skin of different groups was observed and analyzed by pathological technology(immunohistochemical staining).Furthermore,we investigated the viability of skin in five respective groups evaluated using oxygen consumption assay.Results: integrin β1 and talin were affected in different temperature for preservation.In response to lowering of temperature,the content of integrin β1 and talin were increased.The mechanism underlying these activities might involve damage of skin by low temperature and various alterations appeared while the samples were stored at different temperatures.The skin which was preserved at-196 ℃ showed less effect compared with other groups.Conclusion: These results demonstrate that the cryoinjury of skin is correlated with changes of the two proteins,and integrin β1 and talin play important roles in cryopreservation.
