Establishment of human stomach poor-differentiation adenocarcinoma(PdGC-81)cell line and its biological characters

Abstract
The cell line designated as PdGC-81,and derived from poordifferentiation adenocarcinoma of human stomach has been transfered for 83 passages in 13 months. The specimens were obtained from a female patient,65 years of age,on whom a poor-differentiation adenocarcinoma of stomach had been demonstrated by gastroscopy biopsy.The metastatic node of greater omentum was taken.After demonstration as poor-differentiation adenocarcinoma, the specimen was cut into small blocks of 1 mm3,then planted in some small bottles.The primary epithelial cell crown was formed within one week.The first passage was done on the 16th day after cleaning off the fibroblasts.The subculture transfer was made once every 7-10 days. After 5th passage the fibroblast has been cleaned basically.Then the transfer was made once every 4-6 days.The medium was changed 2-3 times weekly. The PdGC-81 cell line showed the following characters:1.The pathological type of the gastroscopy biopsy and the primary planted material were similar to the type of the animal tumor transplanted with PdGC-81 cell line.2.Microscopically,the major cells of the line were presented as elliptic and polygonal epithelial cells with abnormal mitotic figures and 2-4 nucleoli.3. Electron-microscopically,there were rich microvilli on the surface of the cell,and desmosomes were also observed,along with a lot of ribosomes.4.The growth curves of the 33th and 65th passage,were determined.The cells of these passage were proliferated 23 and 21 times respetively within 8 days.The doubling time was 41.4 and 43.4 hrs respectively.The mitotic index attained its peak to as much as 80.5‰.on the 3th day in 33th passage and 72‰.on the 4th day in 65th pasage,5.Chromosome analysis of the PdGC-81 cell line was performed by observing 100 metaphases of mitotic division,finding that the number of chromosome varied widely in different cells,range 56-60.This cell line may be considered as subtriploid.
