Using LDH release assay to detect cytopathic extent and determine antiviral activity of interferons

Objective: Using LDH release assay detect the antiviral activity of Interferons. Methods: On the basis of LDH activity quantified after WISH cells were lysed, cytopathic extent was determined and antiviral activity of interferon detected. LDH release assay was improved and was compared with the crystal violet staining and the MTT colorimetry on sensitivity, repeatability and relateness.Results and Conclusions:The detection procedures, reagent cost and time were reduced in LDH release assay with higher sensitivity and better repeatability. There was no significant difference between LDH release assay, crystal violet stained and MTT colorimetry. LDH release assay was convenient and applicable and was better than crystal violet staining and MTT colorimetry.
