跨膜蛋白Lgr5作为胃肠道恶性肿瘤干细胞分子表面标记物的研究进展

Research advances in transmembrane protein Lgr5 as a molecular surface marker of stem cells from gastrointestinal malignancies

  • 摘要: 肿瘤干细胞具有持续自我更新和多向分化的特性,是导致肿瘤复发和耐药的主要原因。能否准确识别并靶向摧毁肿瘤干细胞,已经成为肿瘤学研究的新方向和热点问题。跨膜蛋白Lgr5广泛表达于消化道组织,具有极强的干细胞活性,并且在消化道恶性肿瘤细胞膜表面特异性高表达。深入了解Lgr5蛋白的分子特点有助于提高消化道肿瘤干细胞分选效率,对进一步研究消化道肿瘤的发生发展机制、探索新的治疗方法、提高总体预后具有重要意义。本文将以Lgr5蛋白结构特点为基础,结合信号转导通路和不同胃肠道肿瘤生物学活性特点,总结追踪近年来Lgr5蛋白作为干细胞标记物在胃肠道肿瘤基础领域的最新进展,为下一步研究提供理论储备。

     

    Abstract: Tumor stem cells have the property of continuous self-renewal and multi-directional differentiation, which are the main causes of tumor recurrence and drug resistance. Whether the tumor stem cells can be accurately identified and targeted for destruction has become a new direction and hot topic in the research of oncology. The transmembrane protein Lgr5 is widely expressed in the digestive tract and has extremely strong stem cell activity. In addition, it has the characteristics of high specific expression on the membrane surface of gastrointestinal malignant tumors. In-depth understanding of the molecular characteristics of Lgr5 protein will help improve the sorting efficiency of gastrointestinal tumor stem cells, which is of great significance for further research on the occurrence and development mechanism of gastrointestinal tumors, exploration of new therapeutic methods, and improvement of the overall prognosis. Based on the structural characteristics of Lgr5 protein, in combination with the signal transduction pathways and the biological activity characteristics of different gastrointestinal tumors, this paper reviews the latest progress of the Lgr5 protein as a stem cell marker in the basic research field of gastrointestinal tumors in recent years, so as to provide theoretical foundation for further research.
