Expression and function of CDK4 gene in osteosarcoma cells

      背景   细胞周期蛋白依赖性激酶4(cyclin dependent kinase 4,CDK4)属于丝氨酸/苏氨酸激酶家族,是许多癌症的潜在治疗靶点,但其在骨肉瘤中的表达水平及功能尚不清楚,有待深入研究。
      目的   本研究旨在探索CDK4基因在骨肉瘤细胞中的表达水平,并验证该基因与骨肉瘤细胞增殖和迁移的关系。
      方法   选择GEO数据库中与骨肉瘤相关的基因表达谱芯片数据,对比数据集内两组样品骨肉瘤MG63细胞系及正常成骨细胞HOB细胞系的数据,分析CDK4基因在两组间的表达差异。使用人乳腺文库作模板,利用聚合酶链反应(polymerase chain reaction,PCR)将人CDK4基因的CDS区编码序列进行扩增得到扩增产物。将扩增产物插入pEGFP-C1载体中,得到重组质粒,再将此重组质粒转染至MG63细胞内,利用蛋白质免疫印迹(Western blot)方法检测质粒的表达情况,并用荧光显微镜观察带有荧光标记的CDK4蛋白在细胞中的定位。通过CCK-8、克隆形成及划痕实验研究转染CDK4重组质粒的人骨肉瘤MG63细胞的增殖和迁移情况。
      结果   基因表达谱芯片数据集分析显示,CDK4在MG63细胞系中的表达水平显著高于HOB细胞系。将获得的质粒进行双酶切和测序,成功构建pEGFP-CDK4真核表达载体;Western blot证明目的蛋白成功表达;荧光显微镜观察发现CDK4定位在细胞核中;CCK-8、克隆形成和划痕实验的结果显示,过表达pEGFP-CDK4能促进MG63细胞的增殖和迁移。
      结论   CDK4在人骨肉瘤MG63细胞系中的表达水平高于正常成骨细胞HOB细胞系。CDK4蛋白定位于MG63细胞核中,可能有助于促进骨肉瘤细胞的增殖和迁移。

     

    Abstract:
      Background   Cyclin dependent kinase 4 (CDK4) belongs to the serine/threonine kinase family, which is a potential target for many cancers. Its expression level and function in osteosarcoma remain unclear and need to be further studied.
      Objective   To investigate the expression level and function of CDK4 gene in osteosarcoma cells and verify its relationship with the proliferation and migration of osteosarcoma cells.
      Methods   The microarray data of gene expression profiles related to osteosarcoma were searched in GEO database. Osteosarcoma MG63 cell line and normal osteoblast HOB cell line in the dataset were compared to analyze the differences of CDK4 gene expression between the two groups. Using the human mammary gland library as a template, polymerase chain reaction (PCR) was carried out to amplify the CDS region coding sequence of human CDK4 gene to obtain the amplification product. Subsequently, the amplification product was inserted into the pEGFP-C1 vector to obtain the recombinant plasmid, which was then transfected into the human osteosarcoma MG63 cell line. The expression of the plasmid was detected by Western blot, and the location of the fluorescent labeled CDK4 protein within cells was observed by fluorescence microscope. The proliferation and migration of MG63 cells transfected with CDK4 recombinant plasmid were studied by CCK-8 assay, clone formation assay and scratch test.
      Results   The gene expression profile microarray dataset analysis showed that the expression of CDK4 in human osteosarcoma MG63 cell line was significantly higher than that in normal osteoblast HOB cell line. The obtained plasmids were digested and sequenced. The results showed that the eukaryotic expression vector pEGFP-CDK4 was successfully constructed. Western blot proved that the target protein was successfully expressed. Fluorescence microscopy showed that CDK4 protein was located in the nucleus of osteosarcoma cells. CCK-8 assay, colony formation assay and scratch test showed that overexpression of pEGFP-CDK4 promoted the proliferation and migration of MG63 cells.
      Conclusion   The expression of CDK4 in human osteosarcoma MG63 cells is significantly higher than that in normal osteoblast HOB cells. CDK4 protein is located in the nucleus of MG63 cells and can probably promote the proliferation and migration of osteosarcoma cells.
