NETosis in heat stroke: From mechanistic insights to future therapeutic strategy

  • 摘要: 热射病是以中枢神经系统损伤及严重高热为主要特征的疾病,可迅速进展为多器官功能不全综合征导致患者死亡,与脓毒症的发病机制类似。中性粒细胞炎性死亡(NETosis)在脓毒症中发挥了核心作用,但其在热射病中的研究尚不深入,也未得到足够的重视。本综述旨在总结NETosis 在热射病及其导致多器官功能不全综合征中的机制研究现状。并探讨靶向NETosis 通路作为潜在治疗策略的科学依据与前景,以期为深入阐明热射病的发病机制和开发新型干预手段提供理论参考。

     

    Abstract: Heatstroke (HS) is a life-threatening condition characterized by central nervous system dysfunction and severe hyperthermia, which can rapidly progress to multiple organ dysfunction syndrome (MODS) and death. The pathogenesis of HS shares similarities with that of sepsis. NETosis, a unique form of neutrophil cell death, plays a central role in sepsis; however, its role in HS remains understudied and underappreciated. This review aims to summarize the current understanding of the role of NETosis in HS and HS-induced MODS. Furthermore, we discuss the scientific rationale and future prospects of targeting the NETosis pathway as a potential therapeutic strategy, with the goal of providing a theoretical foundation for elucidating the pathogenesis of HS and developing novel interventions.
