bFGF对体外培养新生牛RPE细胞非特异性吞噬功能的影响

Stimulative effects of bFGF on the nonspecific phagocytosis of newborn bovine RPE cells in vitro

  • 摘要: 目的: 为探讨碱性成纤维细胞生长因子(bFGF)对视网膜色素上皮(RPE)细胞吞噬功能的影响。方法: 采用新生牛RPE细胞培养,并行免疫组化鉴定。对培养的RPE细胞于0h、24h、48h 3个时间点分别给予0ng/ml、1ng/ml、4ng/ml、16ng/ml的bFGF后,加入鸡红细胞悬液,孵育1h,经清洗固定后于光镜下观察、计数,以判断RPE细胞吞噬功能。结果: 应用bFGF后,RPE细胞吞噬率及吞噬指数均明显增加,在一定范围内呈浓度及时间依赖性。结论: bFGF对牛RPE细胞的非特异性吞噬功能有促进作用。

     

    Abstract: Objective: To observe the effects of bFGF (basic fibroblast growth factor) on the nonspecific phagocytosis of newborn bovine RPE cells in vitro. Methods: Primary newborn bovine RPE cells were cultured in vitro and immunocytochemical staining were performed for cell differentiation. bFGF were used at different concentrations and different durations. Chicken RBC (red blood cell) were used to judge the nonspecific phagocytosis. Results: The phagocytic count and phagocytic rate of RPE were increased by bFGF in concentration and time dependent manner compared with the control group. Conclusion: bFGF can stimulate the bovine RPE cells′nonspecific phagocytosis in vitro.
