钙通道阻断剂硝苯地平对血迷路屏障的通透性研究

Calcium channel blocker (nifedipine): pharmacokinetics in cochlear perilymph of guinea pig

  • 摘要: 目的: 探讨钙通道阻断剂硝苯地平的血迷路屏障通透性,建立高效液相色谱仪测定豚鼠外淋巴液硝苯地平浓度的方法。方法: 豚鼠腹腔注射硝苯地平1周采集外淋巴液,应用反相高效液相色谱法(reversedphasehigh performanceliquidchromatography, RT HPLC)测定外淋巴液硝苯地平的浓度。结果: 本法的灵敏度为5ng/ml。在浓度为50ng/ml时的平均回收率为98%。测定的相对偏差为8. 56%。外淋巴液硝苯地平的浓度为50. 2~124. 2ng/ml。结论: 硝苯地平可以通过血迷路屏障,浓度随给药时间变化。本实验方法简便、快捷、准确可靠,可有效用于豚鼠外淋巴液硝苯地平的药理学研究。

     

    Abstract: Objective:To investigate the determination of nifedipine in perilymph of guinea pig. Methods:The guinea pigs were injected with nifedipine for one week, and then perilymphatic fluids of guinea pig were aspirated at 30min, 150min and 270min after injection. The nifedipine in perilymph was determined by using reversed phase high-performance liquid chromatography (RP-HPLC).Results:The sensitivity introduced in our article is 5ng/ml. The average recovery is 98% at the concentration of 50ng/ml. The relative precision is 8.56%. The concentrations of nifedipine in perilymph of guinea pig range from 50.2 ng/ml to 124.2ng/ml. Conclusions:The study indicates that nifedipine has excellent penetration into perilymph. RP-HPLC can be used effectively in pharmacology determination of nifedipine in perilymph of guinea pig.
