Abstract:
Objective: This work was undertaken to investigate the effect of diazoxide,sodium nitroprusside, methyene blue, indomethacin, L-NAME on aortic function of two-kidney and one-clip rats with renal hypertension. Methods: Under antiseptic conditions, the left renal artery was exposed renal vein through a retroperitoneal flank incision and carefully dissected free of the left. A silver clip with an internal diameter of 0.20.3mm was placed around the left artery, resulting in partial occlusion of renal perfusion. SBP was observed by tail blood pressure. The aortic rings was contracted by norepinephrine at a final concentration of 0.62μmolL
-1 or KCl 60mmolL
-1. Results: NE-induced aortic contractions were reduced by diazoxide, yielding EC
50L
95 values of (27.942.7)μmol/L(NTR) (51.2194.3)μmol/L(RHR), the difference between them was significant((P<0.01)); NE-induced aortic contractions were reduced by sodium nitroprusside, yielding EC
50L
95 values of (0.15(0.14)μmol/L)(NTR) (0.060.63)μmol/L(RHR), the difference between them was not significant(P>0.05).The effects of acetycholine decreased the KCl-induced aortic contraction were different between NTRand RHR, the effect on RHR aortic rings was more poor than on NTR. Conclusion: At renal conditions, diastole of aorta was more poor than at normotensive conditions.
