骨性关节炎患者血清和关节滑液中BMP-7的检测及临床意义

Measurement of bone morphogentic protein 7 level in serum and synovial fluid from patients with osteoarthritis and its clinical significance

  • 摘要: 目的 探讨将BMP-7作为骨性关节炎(osteoarthritis,OA)早期诊断潜在生物学标志的可能性。 方法 收集67例OA患者及23例非OA膝关节疾病患者(半月板损伤11例,前交叉韧带损伤9例,盘状半月板2例,髌骨半脱位1例,均无明显关节软骨损伤)的血清和关节滑液,采用Kellgren-Lawrence放射学分级标准对OA患者的影像学表现分级。酶联免疫吸附试验(ELISA)检测血清和关节滑液中BMP-7水平。 结果 OA组血清、滑液中BMP-7水平高于非OA组(Pser< 0.001,PSF=0.001),两组血清中BMP-7水平均高于滑液(P< 0.001)。OA组血清、滑液中BMP-7水平与OA严重程度相关(rser=0.788,Pser< 0.001;rSF=0.766,PSF< 0.001),血清BMP-7与滑液BMP-7水平呈正相关(r=0.817,P< 0.001)。 结论 BMP-7可作为生物学标志用于OA的早期诊断及预后监测。

     

    Abstract: Objective To study the possibility to use bone morphogenetic protein-7 (BMP-7) as a potential biomarker in early diagnosis of osteoarthritis (OA). Methods Plasma and synovial fluid (SF) samples were taken from 67 OA patients and 23 non-OA patients (11 meniscus injury patients, 9 anterior cruciate ligament injury patients, 2 discoid meniscus patients, and 1 patellar subluxation patient, with no significant articular cartilage lesion). Findings on their images were classified according to the Kellgren-Lawrence radiographic grading criteria. The BMP-7 levels in serum and SF were measured by ELISA. Results The BMP-7 levels in serum and SF were significantly higher in OA patients than in non-OA patients (P< 0.001) and in serum than in SF (P< 0.001), and correlated with the severity of OA (r=0.788, r=0.766, P< 0.001). In addition, the BMP-7 levels in serum and SF were positively correlated with each other (r=0.817, P< 0.001). Conclusion BMP-7 can be used as a biomarker in early diagnosis of OA and its prognosis monitoring.
