木果楝内酯抑制人乳腺癌脑转移细胞增殖活性的研究

Role of xylolactones in inhibiting proliferation of brain metastatic human breast cancer cells

  • 摘要: 目的 研究3种木果楝内酯化合物(Xylocarpin H,Xylogranatin C和Xylomexicanin A)抑制人乳腺癌脑转移细胞增殖的作用及分子机制。 方法 3种木果楝内酯化合物分别处理人乳腺癌脑转移(KT)细胞,MTT比色法检测其对KT细胞增殖活性的抑制作用;采用Western blot技术检测细胞凋亡相关蛋白的表达变化。 结果 MTT比色法显示Xylogranatin C对KT细胞增殖有较强的抑制作用,呈浓度依赖性;Western blot结果显示Xylogranatin C处理的KT细胞p53、Bax和Caspase-3蛋白表达增强,与对照组比较有统计学意义。 结论 木果楝内酯化合物Xylogranatin C具有较强的抑制人乳腺癌脑转移细胞增殖的作用,机制与诱导肿瘤细胞凋亡相关。

     

    Abstract: Objective To study the role of xylocarpin H, xylogranatin C and xylomexicanin A in inhibiting proliferation of brain metastatic human breast cancer cells (KT cells) and their molecular mechanisms. Methods KT cells were treated with xylocarpins H, C and A. Inhibitory effects of xylocarpins H, C and A on proliferation of KT cells were detected by MTT assay. Apoptosis-related protein expression level in KT cells was measured by Western blot. Results MTT assay showed that xylogranatin C inhibited the proliferation of KT cells in a dose-dependent manner. Western blot displayed that the expression levels of p53, Bax and caspase-3 were significantly higher in xylogranatin C-treated KT cells than in controls (P< 0.01, P< 0.05). Conclusion Xylogranatin C can effectively inhibit the proliferation of KT cells by inducing their apoptosis.
