正加速度暴露后大鼠心室肌LC3发生高表达变化

Over-expression of LC3 in ventricular muscle of rats after exposure to +Gz

  • 摘要: 目的 探讨正加速度(+Gz)暴露后大鼠心室肌自噬相关基因微管相关蛋白轻链3(LC3)的表达变化。 方法 雄性SD大鼠18只,随机分为对照组和+10 Gz/5 min暴露组。分别于+Gz暴露后6 h、24 h取心室肌组织,采用RT-PCR和Western blot方法测定LC3 mRNA及蛋白表达的变化。 结果 与对照组相比,+Gz暴露后6 h心室肌LC3 mRNA及蛋白表达均显著增强(P<0.05),暴露后24 h其表达量仍显著增加(P<0.05)。 结论 +10 Gz/5 min暴露可诱导大鼠心室肌自噬相关基因LC3的高表达,提示高+Gz可能引起了心室肌自噬的激活。

     

    Abstract: Objective To study the expression of autophagy-related marker LC3 in ventricular muscle of rats after exposure to +Gz. Methods Eighteen male SD rats were randomly divided into control group and +10 Gz/5 min group. Their ventricular muscle tissue samples were taken at 6 and 24 h after exposure to +10 Gz. Expressions of LC3 mRNA and protein were detected by RT-PCR and Western blot, respectively. Results The expression levels of LC3 mRNA and protein were significantly higher in +10 Gz/5 min group than in control group at 6 and 24 h after exposure to +Gz (P< 0.05). Conclusion Exposure to +10 Gz can induce overexpression of LC3 in ventricular muscle of rats by activating the autophagocytic function of their ventricular muscle.
