慢性睡眠限制对幼鼠空间学习记忆能力的影响

Effect of chronic sleep restriction on spatial learning and memory in infantile rats

  • 摘要: 目的 探讨慢性睡眠限制对幼鼠空间学习记忆能力的影响。 方法 将24只1月龄SD雄性鼠随机分为慢性睡眠限制组、睡眠剥夺组和对照组,每组8只。对慢性睡眠限制组幼鼠进行连续5 d每天6 h的睡眠限制,采用“温和处理法”;对睡眠剥夺组幼鼠进行连续5 d每天6 h的睡眠剥夺,采用“多平台水环境法”。分别在睡眠干预前和干预第1、3、5天应用Morris水迷宫方法检测幼鼠的空间学习记忆能力。 结果 水迷宫实验显示睡眠限制组、睡眠剥夺组幼鼠在睡眠干预的第3天、第5天,到达平台的平均潜伏期、探索路程较对照组延长(P< 0.05),其中睡眠剥夺组在第5天的影响最显著(P< 0.01);在睡眠干预的第5天,睡眠限制组、睡眠剥夺组幼鼠之间出现了差别,睡眠剥夺组幼鼠的潜伏期、探索路程较睡眠限制组延长(P< 0.05);而在空间探测实验中,3组幼鼠在平台所在象限时间的百分比、穿环次数有显著差异:睡眠剥夺组较睡眠限制组减少(P< 0.05),较对照组显著减少(P< 0.01);睡眠限制组较对照组减少(P< 0.05)。 结论 连续5 d、每天6 h的慢性睡眠限制可减弱幼鼠的空间学习记忆能力,但其影响程度小于同等时间的睡眠剥夺。

     

    Abstract: Objective To study the effect of chronic sleep restriction (CSR) on spatial learning and memory ability in infantile rats. Methods Twenty-four male infantile SD rats at the age 1 month were randomly divided into CSR group, sleep deprivation (SD) group, and control group (8 in each group). Rats in CSR group and SD group underwent sleep restriction by gentle handing and multiple water platform techniques, respectively, 6 h a day for 5 days. Their spatial learning and memory ability were assessed by Morris water maze test before and 1, 3, 5 days after sleep intervention. Results Morris water maze test showed that the average latent period and distance were significantly longer in CSR group and SD group than in control group on days 3 and 5 after sleep intervention (P< 0.05), especially in SD group than in control group on day 5 after sleep intervention (P< 0.01). The latent period and distance were significantly longer in SD group than in CSR group on day 5 after sleep intervention (P< 0.05). Spatial probe test showed that the time in target quadrant (TTQ) and the number of platform crossings (NPC) were significantly lower in SD group than in CSR group and control group (P< 0.05). Conclusion CSR (6 h a day for 5 days) impairs the spatial learning and memory ability in infantile rats. However, its effect is ≤ that of sleep deprivation on the spatial learning and memory ability in infantile rats.
