热打击对大鼠肺组织血管内皮生长因子A的影响

Heat stress on vascular endothelial growth factor A in lungs of rats

  • 摘要: 目的 研究热打击对大鼠肺组织血管内皮生长因子A(vascular endothelial growth factor A,VEGF-A)及相关细胞因子的影响。 方法 雄性SD大鼠30只,随机分为空白对照组(n=10),热打击+恢复15 min组(n=10),热打击+恢复3 h组(n=10)。热打击组大鼠麻醉后置于高温高湿模拟环境中,肛温达到42℃后置于室温分别恢复15 min及3 h。酶联免疫法(Elisa)测各组大鼠肺组织匀浆VEGF-A及相关细胞因子水平。 结果 热打击+恢复15 min组VEGF-A、转化生长因子β1(transforming growth factor beta1,TGF-β1)水平较空白对照组显著升高,热打击+恢复3 h组VEGF-A、TGF-β1水平较空白对照组显著降低,且VEGF-A水平与TGF-β1水平呈直线回归关系。热打击+恢复3 h组促炎细胞因子TNF-α水平略高于热打击+恢复15 min组和空白对照组。3组大鼠促炎细胞因子IL-6水平差异无统计学意义。 结论 热打击早期大鼠肺组织VEGF-A水平短暂升高后,下降至正常水平以下,可能成为热打击后大鼠急性肺损伤发病初始阶段的重要环节,且该变化可能仅与热直接损伤有关。

     

    Abstract: Objective To investigate the effect of heat stress on vascular endothelial growth factor A and related molecules in lungs of rats. Methods Thirty male SD rats were randomly divided into three groups: non-HS group (n=10), HS + 15-minute-recovery group (n=10) and HS + 3-hour-recovery group (n=10).Two heated groups of animals were put in a folded heating pad with general anesthesia. Once their core temperature (Tc) elevated to 42℃, the heated animals were allowed to recover at a room temperature for 15 min and 3 h. Levels of vascular endothelial growth factor A (VEGF-A) and related molecules of the lung homogenate were detected by enzyme-linked immunosorbent assay. Results Compared with the non-HS group, the levels of VEGF-A and transforming growth factor beta1 (TGF-β1) increased significantly in the HS + 15-minute-recovery group, while decreased noticeably in the HS + 3-hour-recovery group. The level of pre-inflamatory cytokine TNF-α slightly increased in the HS + 3-hour-recovery group. However, no significant difference of the pre-inflamatory cytokine IL-6 was found among the three groups. Conclusion The level of VEGF-A which may be only affected by heat injury in the early stage of heat stress decreases below the normal level after a transient elevation, and it plays a crucial role in the development of acute lung injury (ALI) in rats under heat stress.
