脂肪间充质干细胞对巨噬细胞清除凋亡中性粒细胞的影响

Effect of adipose-derived mesenchymal stem cells on macrophage phagocytosis of apoptotic neutrophils

  • 摘要: 目的 探讨脂肪间充质干细胞(adipose-derived mesenchymal stem cell,ADSC)对巨噬细胞极化及其对凋亡中性粒细胞吞噬能力的影响,为炎症的干细胞治疗提供新的依据。 方法 体外分离、培养、鉴定大鼠脂肪间充质干细胞、巨噬细胞和中性粒细胞,荧光染料CFSE标记中性粒细胞,分为巨噬细胞、巨噬细胞+ADSC、巨噬细胞+ADSC上清液、巨噬细胞+中性粒细胞、巨噬细胞+ADSC+中性粒细胞、巨噬细胞+ADSC上清液+中性粒细胞 6组进行细胞共培养实验,通过免疫荧光染色和流式细胞学检测细胞表型变化和吞噬能力。 结果 与巨噬细胞组相比,巨噬细胞+ADSC、巨噬细胞+ADSC组细胞CD163阳性率明显增高;与巨噬细胞+中性粒细胞相比,巨噬细胞+ADSC+中性粒细胞、巨噬细胞+ADSC上清液+中性粒细胞组细胞CFSE阳性率明显增高,CFSE+CD163+的细胞明显增加。 结论 ADSC通过促进巨噬细胞向M2c型极化,增强巨噬细胞对凋亡中性粒细胞的吞噬能力。

     

    Abstract: Objective To investigate the effect of adipose-derived mesenchymal stem cell (ADSC) on macrophage polarization and macrophage phagocytosis of apoptotic neutrophils. Methods Rat ADSC, macrophages and neutrophils were isolated, cultured, and identified. neutrophils were labeled with CFSE fluorescent dye. Cells were co-cultured accordingly: macrophage, macrophage+ADSC, macrophage+ADSC supernatant, macrophage+neutrophil, macrophage+ADSC+neutrophil, macrophage+ADSC supernatant+neutrophil. Polarization and phagocytosis were measured using immunofluorescence staining and flow cymetory analysis. Results Compared with macrophage group, percentage of CD163+cells increased significantly in macrophage+ADSC group and macrophage+ADSC supernatant group. Compared with macrophage+neutrophil group, percentages of CD163+cells and CFSE+CD163+cells significantly increased in macrophage+ADSC+neutrophil group and macrophage+ADSC supernatant+neutrophil group. Conclusion ADSC facilitates macrophage phagocytosis of apoptotic neutrophils through turning macrophages into M2c type.
