子宫内膜异位症患者内膜中miR-20b-5p、miR-106a-5p的表达及机制探讨

Expression and role of miR-20b-5p and miR-106a-5p in patients with endometriosis

  • 摘要: 目的 检测miR-20b-5p、miR-106a-5p、PTEN mRNA、CCNE1 mRNA在正常子宫内膜(N)、子宫内膜异位症(endometriosis,EM)患者在位内膜(eutopic endometrial,Eu)、子宫内膜异位症患者异位内膜(ectopic endometrial,Ec)组织中的表达,并探讨miR-20b-5p、miR-106a-5p对子宫内膜异位症细胞增殖与凋亡的影响。 方法 选择2017年5月- 2018年5月在本院妇产科行腹腔镜手术及术后病理检查确诊的EM患者30例,年龄(32.4±6.2)岁,对照组选择同期行腹腔镜子宫肌瘤剔除术的患者30例,年龄(36.35±5.89)岁。采用qRT-PCR法检测Eu(n=30)、Ec(n=37)以及N(n=30)组织中miR-20b-5p、miR-106a-5p、PTEN mRNA、CCNE1 mRNA的表达,并用Pearson相关检验分析miR-20b-5p、miR-106a-5p与PTEN mRNA、CCNE1 mRNA的相关性。 结果 Ec与Eu相比,miR-20b-5p、miR-106a-5p表达下降(P<0.001),PTEN mRNA的表达上升(P<0.001),CCNE1 mRNA的表达下降(P=0.005);Eu与N相比,miR-20b-5p表达上升(P=0.038),PTEN表达下降(P=0.049),miR-106a-5p及CCNE1的表达未见明显差异(P>0.05);miR-20b-5p与PTEN mRNA在Eu及Ec中的表达呈负相关(r=-0.63,P<0.001;r=-0.42,P=0.01);miR-106a-5p与PTEN mRNA的表达在Eu(r=-0.14,P=0.465)及Ec(r=-0.25,P=0.142)中不存在相关性。 结论 miR-20b-5p可能通过PTEN上调细胞增殖,促进EM的发病。

     

    Abstract: Objective To explore the expression of miR-20b-5p, miR-106a-5p, PTEN mRNA and CCNE1 mRNA in normal endometria (N), eutopic endometria (Eu) and ectopic endometria (Ec), and investigate the role of miR-20b-5p, miR-106a-5p on proliferation and apoptosis of cells in endometriosis. Methods Thirty patients with endometriosis who underwent laparoscopic surgical procedures in our hospital from May 2017 to May 2018 were enrolled in our study. And another 30 patients underwent laparoscopic myomectomy were collected as the controls. The expression of miR-20b-5p, miR-106a-5p, PTEN mRNA, CCNE1 mRNA in Eu (n=30), Ec (n=37) and N (n=30) were detected by qRT-PCR, and the correlation between miR-20b-5p, miR-106a-5p and PTEN mRNA, CCNE1 mRNA was analyzed by Pearson correlation analysis. Results Compared to Eu, the levels of miR-20b-5p and miR-106a-5p were significantly lower in Ec (P< 0.001), and the expression level of PTEN mRNA increased (P< 0.001), while the level of CCNE1 decreased (P=0.005). Compared with N, the expression level of miR-20b-5p was higher in Eu (P=0.038), while the level of PTEN was lower (P=0.049), and there was no statistically significant difference in miR-106a-5p and CCNE1 (P> 0.05, respectively). The level of miR-20b-5p was negatively correlated with PTEN in Eu and Ec group (r=-0.63, P< 0.001; r=-0.42, P< 0.001). There was no correlation between miR-106a-5p and PTEN in both Eu and Ec group (r=-0.14, P=0.465; r=-0.25, P=0.14). Conclusion MiR-20b-5p may promote cell proliferation in endometriosis by regulating PTEN.
