Abstract:
Objective To investigate the expression of CD25 in cancer tissue, paracancerous region tissue and normal gastric tissue, and explore the features and molecular markers that predict the prognosis of patients with resected gastric cancer(GC).
Methods Forty one GC cases at stage Ⅰ-Ⅲ who underwent surgery in Chinese PLA General Hospital from January 2010 to June2011 were enrolled in our study. The samples, including cancer tissue, paracancerous region tissue and normal gastric tissue were collected. The expression of CD25 protein was detected using a two-step immunohistochemical procedure with the EnVision FLEX detection system(DAKO). The patients were followed up till June 30, 2015, and influencing factors of disease free survival(DFS)was analyzed with univariate and multivariate analysis.
Results CD25 was detected with a positive rate of 80.49%, 68.29% and80.49% in cancer tissue, paracancerous region tissue and normal gastric tissue, respectively. Univariate analysis showed that patients with CD25 positive expression had shorter DFS(16 months
vs 48 months,
HR=0.013). And multivariate analysis showed that TNM stage(HR, 11.268; 95% CI, 1.869-67.944), tumor differentiation(HR, 0.026; 95% CI, 0.002-0.334), Lauren typing(HR, 13.558;95% CI, 1.231-149.294), and positive expression of CD25 in cancer tissues(HR, 13.043; 95% CI, 3.107-54.758) were prognostic factors.
Conclusion Positive expression of CD25 can be detected in cancer tissues, paracancerous regions and normal gastric tissues.Patients with positive expression of CD25 have higher risk of recurrence and metastasis and poorer prognosis.
