Abstract:
Objective To explore the relationship between expression of transient receptor potential canonical 6(TRPC6) in human umbilical artery smooth muscle cells(HUASMCs) and severe preeclampsia, and provide clue for exploring a new treatment.
Methods From April to November in 2018, 10 healthy pregnant women and 10 pregnant women with severe preeclampsia who had vaginal delivery or cesarean section in our hospital were selected as control group and experimental group, respectively. The umbilical artery vessels were collected and real-time fluorescent quantitative PCR experiment and western blotting were applied to detect the expression of TRPC6.
Results The relative mRNA expression of TRPC6 in the human umbilical arterial smooth muscle cells of 10 healthy pregnant woman was 9.583±0.913, and it was 8.447±0.982 in pregnant women with severe preeclampsia, with statistically significant difference(
P=0.003). The relative protein expression of TRPC6 in the human umbilical arterial smooth muscle cells of 10 healthy pregnant woman was 0.482±0.081, and it was 0.872±0.079 in pregnant women with severe preeclampsia, with statistically significant difference(
P< 0.001).
Conclusion The mRNA and the protein expression level of TRPC6 in the severe preeclampsia patients are higher than those of the healthy pregnant woman, which suggests that the occurrence of the severe preeclampsia may be related to the up-regulation of expression of TRPC6.
