重度子痫前期孕妇脐动脉血管平滑肌中经典瞬时受体电位通道6的表达

Expression of transient receptor potential canonical 6 in human umbilical artery smooth muscle cells in pregnant woman with severe preeclampsia

  • 摘要: 目的 探索人脐动脉平滑肌细胞(human umbilical arterial smooth muscle cells,HUASMCs)中瞬时受体电位通道6 (transient receptor potential canonical 6,TRPC6)与重度子痫前期的关系,为发现新型治疗方法提供基础理论依据。方法 选取西南医科大学附属医院2018年4-11月产科住院病人经阴道分娩或剖宫产分娩的10例重度子痫前期孕妇为病例组,并匹配同期产科住院的10例健康产妇作为对照组;取分娩后的任意段脐动脉血管,采用实时荧光定量PCR(real-time fluorescent quantitative PCR experiment,QPCR)与蛋白免疫印迹技术(Western blotting)测量TRPC6的表达。结果 QPCR结果显示,正常孕妇组HUASMCs的TRPC6相对表达量为9.583±0.913,重度子痫前期孕妇组HUASMCs的TRPC6相对表达量为8.447±0.982,两组差异有统计学意义(P=0.003)。Western blotting结果也显示正常孕妇组HUASMCs的TRPC6表达显著低于重度子痫前期孕妇组(P<0.001)。结论 重度子痫前期孕妇组HUASMCs中TRPC6的mRNA及蛋白表达水平均高于正常孕妇组,提示重度子痫前期的发病可能与TRPC6的表达上调相关。

     

    Abstract: Objective To explore the relationship between expression of transient receptor potential canonical 6(TRPC6) in human umbilical artery smooth muscle cells(HUASMCs) and severe preeclampsia, and provide clue for exploring a new treatment. Methods From April to November in 2018, 10 healthy pregnant women and 10 pregnant women with severe preeclampsia who had vaginal delivery or cesarean section in our hospital were selected as control group and experimental group, respectively. The umbilical artery vessels were collected and real-time fluorescent quantitative PCR experiment and western blotting were applied to detect the expression of TRPC6. Results The relative mRNA expression of TRPC6 in the human umbilical arterial smooth muscle cells of 10 healthy pregnant woman was 9.583±0.913, and it was 8.447±0.982 in pregnant women with severe preeclampsia, with statistically significant difference(P=0.003). The relative protein expression of TRPC6 in the human umbilical arterial smooth muscle cells of 10 healthy pregnant woman was 0.482±0.081, and it was 0.872±0.079 in pregnant women with severe preeclampsia, with statistically significant difference(P< 0.001). Conclusion The mRNA and the protein expression level of TRPC6 in the severe preeclampsia patients are higher than those of the healthy pregnant woman, which suggests that the occurrence of the severe preeclampsia may be related to the up-regulation of expression of TRPC6.
