miRNA-29b及血管内皮生长因子在子痫前期病程中的作用机制

Changes and relationship of miRNA-29b and VEGF in preeclampsia

  • 摘要: 目的 探讨子痫前期患者microRNA-29b(miR-29b)与血管内皮生长因子A(vascular endothelial growth factor A,VEGF-A)的关系及二者对子痫前期发生发展的影响。方法 收集2018年9月-2019年1月在安徽省妇幼保健院危重症产科住院的22例早发型子痫前期(early onset preeclampsia,EOPE)患者和18例晚发型子痫前期(late onset preeclampsia,LOPE)患者的胎盘组织,20例同期在我院分娩的正常妊娠孕妇胎盘为对照组。采用RT-qPCR法检测胎盘中miR-29b表达,采用免疫组化法检测胎盘组织中VEGF-A蛋白表达,分析二者的关系及对病程的影响。结果 miR-29b在早发型子痫前期、晚发型子痫前期和对照组胎盘中表达量依次降低(3.72±0.56,2.41±0.40和1.46±0.47),而VEGF-A蛋白表达水平在三组间依次升高(0.74±0.05,0.49±0.03和0.29±0.02),组间差异均有统计学意义(all P<0.05)。子痫前期患者miR-29b表达量与VEGF-A蛋白表达量呈负相关(r=-0.691,P=0.00);胎盘中miR-29b表达与子痫前期病情严重程度呈正相关,与发病孕周呈负相关(P<0.05);胎盘VEGF-A蛋白与子痫前期病情严重程度呈负相关,与发病孕周呈正相关(P<0.05)。结论 子痫前期患者miR-29b升高导致血管内皮生长因子A表达降低,可能参与了子痫前期的发生。

     

    Abstract: Objective To investigate the relationship between microRNA-29b (miR-29b) and vascular endothelial growth factor A (VEGF-A),and their effects on the occurrence and development of preeclampsia. Methods From September 2018 to January 2019,we collected placentas from 22 cases of early onset preeclampsia (EOPE),18 cases of late onset preeclampsia (LOPE) admitted to the department of critical care obstetrics,Anhui Provincial Maternal and Child Health Hospital.Another 20 cases of normal pregnant women delivered in our hospital served as control group.The expression of miR-29b in placenta was detected by RT-qPCR,and the expression of VEGF-A protein in placenta was detected by immunohistochemistry,the relationship between miR-29b and VEGF-A and their effects on the course of this disease were analyzed. Results Compared with the control group,the expression of miR-29b in the placenta of patients in the early-onset preeclampsia group and late-onset preeclampsia group was significantly higher (3.72±0.56 and 2.41±0.40 vs 1.46±0.47,P<0.05,respectively),while and the expression of VEGF-A protein was significantly lower (0.29±0.02 and 0.49±0.03 vs 0.74±0.05,P<0.05,respectively).Compared with the late-onset preeclampsia group (2.41±0.40),the expression of miRNA-29b in early-onset preeclampsia group (3.72±0.56) was significantly higher (P=0.001),while the expression of VEGF-A (0.29±0.02) was significantly lower (P<0.05).There was a negative correlation between the expression of miR-29b and the expression of VEGF-A protein in preeclampsia patients (r=-0.691,P=0.00).The expression of miR-29b in the placenta was positively correlated with the severity of preeclampsia,and negatively correlated with the gestational age (all P<0.05).However,the expression of VEGF-A protein in the placenta was negatively correlated with the severity of preeclampsia,and positively correlated with the gestational age (P<0.05). Conclusion Increased miR-29b in preeclampsia patients leads to decreased expression of vascular endothelial growth factor,which might affect the development of preeclampsia.
