外泌体改造和修饰研究进展

Research advances in modification of exosomes

  • 摘要: 外泌体参与细胞通讯等过程,具有毒性低、免疫原性低、生物相容性高、可穿过血脑屏障等特点,可作为药物载体发挥治疗作用。为避免被免疫系统快速清除、提升靶向细胞特异性、提高细胞内传递效率,需要对外泌体进行改造和修饰。可通过"细胞工程"转染蛋白质或寡核苷酸装载到外泌体;也可通过"外泌体工程"表面改造或修饰功能配体,增强外泌体的药物载体系统的作用。此外,外泌体配体的功能化可以减缓外泌体的清除和跨膜效率。本文综述了外泌体改造和修饰方法的进展,探讨功能化外泌体的改进策略。

     

    Abstract: Exosomes are involved in cell communication and other processes,with the characteristics of low toxicity,low immunogenicity,high biocompatibility,and capability of crossing the blood-brain barrier.However,exosomes need to be modified to avoid rapid clearance by the immune system,enhance target cell specificity,and improve intracellular delivery efficiency.In recent years,transfection of proteins or oligonucleotides into exosomes by "cell engineering" or modification of ligands by "exosome engineering" can enhance the role of the drug carrier system of exosomes.In addition,functionalized exosome ligands can slow exosome clearance and transmembrane efficiency.This article reviews the progress of exosome modification methods and explores improved strategies for functionalized exosomes.
